SCIENTIFIC PAPERS - High-Throughput Drug Screening of the DPC4 Tumor-Suppressor Pathway in Human Pancreatic Cancer Cells - DPC4 (deleted in pancreatic carcinoma, locus 4 or MADH4/SMAD4) is a tumor-suppressor gene mutated in approximately half of human pancreatic adenocarcinomas. The current study reports the use of a stably integrated DPC4-reporter system in human pancreatic cells to screen a library of 16,320 random compounds, searching for chemicals that interact with the DPC4 tumor-suppressive pathway. The screening identified several compounds capable of augmenting and inhibiting DPC4-specific luciferase reporter activity. The identification of such compounds will enable us to further understand this critical signal transduction pathway and may identify potential candidates for rational therapeutic intervention./
