Basic Science - 2000 Volvo Award Winner in Basic Science Studies - Exogenous Tumor Necrosis Factor-Alpha Mimics Nucleus Pulposus-Induced Neuropathology: Molecular, Histologic, and Behavioral Comparisons in Rats - The molecular form and activity of TNFa in herniated nucleus pulposus were determined and compared with that of a biologically active, exogenous 17-kDa form of TNFa applied to rat nerve roots. Both herniated nucleus pulposus and exogenous TNFa produced similar changes in nerve root pathology and pain, measured as mechanical allodynia. The data substantiate the importance of inflammation in the pathogenesis of sciatica./

Basic Science - An Immunohistochemical Study of the Dorsal Capsule of the Lumbar and Thoracic Facet Joints - Thoracic and lumbar joint capsules were analyzed immunohistochemically. Fibrocartilage differentiation of the capsules in the lumbar region suggests they function as 'wrap-around' ligaments that bend around the inferior articular processes as axial rotation occurs to limit further movement./

Basic Science - Correlation Between the Age of Pinealectomy and the Development of Scoliosis in Chickens - Scoliosis developed in 60-80% of chickens pinealectomized within 18 days of hatching, without being influenced by the age at which pinealectomy was performed. Because the serum melatonin levels after pinealectomy were close to zero, yet scoliosis did not develop in all pinealectomized chickens, this study suggests that melatonin plays a complicated role in spinal development. A defect in melatonin synthesis or metabolism contributes to the development of scoliosis in chickens, altbough the detailed role of melatonin is not known./

Basic Science - Direct Current Electrical Stimulation Increases the Fusion Rate of Spinal Fusion Cages - Direct current (DC) stimulation of interbody fusion cages was evaluated in a sheep model. A minimum of seven sheep each received a BAK cage loaded with autograft at L4-L5 with currents of 0, 40, and 100 iA applied by a DC stimulator. DC stimulation increased the histologic and biomechanical fusion rate and the speed of healing of the cages at 4 months./

Basic Science - Dose-Dependent Toxicity of a Commercially Available Demineralized Bone Matrix Material - A survival assay was conducted in athymic nude rats to evaluate the possible dose-dependent adverse effects of a commercially available demineralized bone matrix. All of the eight rats died shortly after an upper bind limb muscle implantation of high-dose Grafton (0.008 mL/g), half of the rats died after a medium dose (0.004 mL/g), and no rats died after a low dose (0.002 mL/g)./

Basic Science - Effects of Anulus Fibrosus and Experimentally Degenerated Nucleus Pulposus on Nerve Root Conduction Velocity: Relevance of Previous Experimental Investigations Using Normal Nucleus Pulposus - Local application of anulus fibrosus to nerve roots had no effect on the nerve conduction velocity, and degenerated nucleus pulposus does not seem to aggravate nucleus pulposus-induced nerve root injury. The use of normal nucleus pulposus therefore may be relevant for evaluating the pathophysiologic mechanisms of nucleus pulposus-induced nerve root injury./
