LABORATORY INVESTIGATIONS - Effects of Isoflurane, Sevoflurane, and Halothane on Myofilament Ca2+ Sensitivity and Sarcoplasmic Reticulum Ca2+ Release in Rat Ventricular Myocytes - Depressed myofilament Ca2+ sensitivity and decreased sarcoplasmic reticulum (SR) Ca2+ content contribute to the negative inotropy of isoflurane and halothane. Sevoflurane does not appear to depress myofilament Ca2+ sensitivity or SR Ca2+ content but decreases the fraction of SR Ca2+ released; this accounts for the negative inotropic effect of sevoflurane./

LABORATORY INVESTIGATIONS - Effects of the Anesthetic Gases Xenon, Halothane, and Isoflurane on Calcium and Potassium Currents in Human Atrial Cardiomyocytes - In isolated human cardiomyocytes, the effects of halothane, isoflurane, and xenon were studied on whole cell currents with the patch clamp technique. Xenon did slightly depress transient outward potassium currents but not L-type calcium currents, whereas halothane and isoflurane exerted stronger inhibitions on potassium currents and depressed calcium currents, in line with the cardiac side effects of the three anesthetics./

LABORATORY INVESTIGATIONS - Effects of Volatile Anesthetics on N-methyl-D-aspartate Excitotoxicity in Primary Rat Neuronal-Glial Cultures - Halothane, sevoflurane, and isoflurane similarly reduced excitotoxic cell death in a dose-dependent manner. This was associated with a reduction in Ca+ + uptake and preservation of mitochondrial membrane potential. Inhibition of N-methyl-D-aspartate (NMDA) toxicity at clinically relevant concentrations of volatile anesthetic was minimal and markedly less potent than that obtained from selective NMDA receptor antagonism./

LABORATORY INVESTIGATIONS - Enflurane Actions on Spinal Cords from Mice That Lack the b3 Subunit of the GABAA Receptor - Deletion of the GABAA receptor b3 subunit did not alter the sensitivity of mouse spinal cord responses to enflurane, but did reduce the role of GABAA receptors in mediating the actions of enflurane. This mutation thus resulted in a quantitative shift in the molecular basis for enflurane depression of spinal neurotransmission, in which other anesthetic target sites replaced GABAA receptors./

LABORATORY INVESTIGATIONS - Enflurane Directly Depresses Glutamate AMPA and NMDA Currents in Mouse Spinal Cord Motor Neurons Independent of Actions on GABAA or Glycine Receptors - The results of the current study show a direct depressant effect of enflurane on glutamate responses in spinal cord motor neurons. Depression persists if inhibitory receptors are blocked. These results are consistent with the hypothesis that immobility as an anesthetic endpoint results from actions on motor neurons, and that direct depression of glutamate excitatory responses plays a role independent of enhancement of g-aminobutyric acid AA or glycine inhibition./

LABORATORY INVESTIGATIONS - Facilitated Uptake of Fentanyl, but Not Alfentanil, by Human Pulmonary Endothelial Cells - Saturable, facilitated fentanyl uptake was observed in human lung microvascular endothelial cells and was inhibited by verapamil but was unaffected by a blocking antibody to P-glycoprotein. Alfentanil uptake by these cells occurred by diffusion alone. These results suggest that specific uptake and sequestration of fentanyl by human lung microvascular endothelial cells may be the mechanisms of its extensive pulmonary uptake/
