LABORATORY INVESTIGATIONS - Enflurane Actions on Spinal Cords from Mice That Lack the b3 Subunit of the GABAA Receptor - Deletion of the GABAA receptor b3 subunit did not alter the sensitivity of mouse spinal cord responses to enflurane, but did reduce the role of GABAA receptors in mediating the actions of enflurane. This mutation thus resulted in a quantitative shift in the molecular basis for enflurane depression of spinal neurotransmission, in which other anesthetic target sites replaced GABAA receptors./

LABORATORY INVESTIGATIONS - Enflurane Directly Depresses Glutamate AMPA and NMDA Currents in Mouse Spinal Cord Motor Neurons Independent of Actions on GABAA or Glycine Receptors - The results of the current study show a direct depressant effect of enflurane on glutamate responses in spinal cord motor neurons. Depression persists if inhibitory receptors are blocked. These results are consistent with the hypothesis that immobility as an anesthetic endpoint results from actions on motor neurons, and that direct depression of glutamate excitatory responses plays a role independent of enhancement of g-aminobutyric acid AA or glycine inhibition./
