PAIN MEDICINE - An Isobolographic Analysis of the Adrenergic Modulation of Diclofenac Antinociception - IMPLICATIONS: Diclofenac analgesia in mice can be modulated by interaction with adrenergic drugs. The systemic but not the intrathecal administration of phenylephrine and clonidine produced supraadditive interactions. For desipramine, prazosin, and yohimbine, supraadditive interactions were not statistically demonstrated. The coadministration of drugs inducing supraadditive effects could be clinically relevant for the treatment of chronic pain because of reduction of doses and side effects./

PAIN MEDICINE - A Prostaglandin E2 Receptor Subtype EP1, Receptor Antagonist (ONO-8711) Reduces Hyperalgesia, Allodynia, and C-fos Gene Expression in Rats with Chronic Nerve Constriction - IMPLICATIONS: We examined whether a prostaglandin E2 receptor subtype EP1, receptor antagonist abrogates neuropathic pain induced by chronic constriction injury model in rats. The EP1 receptor antagonist significantly reduced hyperalgasia, allodynia, and c-fos positive cells. These findings suggested that EP1 receptor antagonists may have a role in treatment of neuropathic pain./

PAIN MEDICINE - The Injectable Cyclooxygenase-2-Specific Inhibitor Parecoxib Sodium Has Analgesic Efficacy When Administered Preoperatively - IMPLICATIONS: Preoperatively administered anti inflammatory analgesics should theoretically prevent the upregulation of nociception and inhibit inflammation and pain by reducing the synthesis of prostaglandins in response to surgery. The findings of this study demonstrate that preoperative administration of parecoxib sodium is both safe and effective for the treatment of pain after oral surgery./

PAIN MEDICINE - The Release of Spinal Prostaglandin E2 and the Effect of Nitric Oxide Synthetase Inhibition During Strychnine-Induced Allodynia - IMPLICATIONS: In a rodent model of allodynia, where pain is triggered by nonpainful stimuli, hair deflection evokes release of spinal prostaglandin E2, locus coeruleus activation, and a blood pressure increase. A mediator role of nitric oxide is also implicated, helping to explain the pathophysiology of this allodynic pain./

PAIN MEDICINE - The Synergistic Antinociceptive Interactions of Endomorphin-1 with Dexmedetomidine and/or S(+)-Ketamine in Rats - IMPLICATIONS: The coadministration of the N-methyl-D-aspartate antagonist receptor antagonist, S(+)-ketamine, or the specific a2-adrenoceptor agonist, dexmedetomidine, significantly enhances the antinociceptive effect of the endogenous m-opioid agonist, endomorphin-1, at the spinal level.
