ANESTHETIC PHARMACOLOGY - (international Society for Anaesthetic Pharmacology) - The Concentration-Effect Relationship of the Respiratory Depressant Effects of Alfentanil and Fentanyl - IMPLICATIONS: This double-blinded, randomized study evaluated the potency ratio of alfentanil and fentanyl-induced respiratory depression. The findings support the notion of parallel analgesic and respiratory depressant effects of alfentanil and fentanyl. Therefore equianalgesic concentrations of both drugs will lead to equally pronounced respiratory depression./

ANESTHETIC PHARMACOLOGY - (international Society for Anaesthetic Pharmacology) - The Convulsant and Anesthetic Properties of CisTrans Isomers of 1,2-Dichlorohexafluorocyclobutane and 1,2-Dichloroethylene - IMPLICATIONS: Cis-trans isomerism can influence the convulsant properties of the nonimmobilizer 2N(1,2-dichlorohexafluorocyclobutane) and the anesthetic properties of dichloroethylene. Such isomeric effects may be as useful as those found with optical isomers in defining receptoranesthetic interactions./

ANESTHETIC PHARMACOLOGY - (International Society for Anaesthetic Pharmacology) - The Determinants of Propofol Induction of Anesthesia Dose - IMPLICATIONS: This study demonstrates a significant relationship between cardiac output and the hypnotic dose of propofol. We suggest that anesthesiologists should include cardiac output, as well as age and weight, in calculating the induction dose of propofol./

ANESTHETIC PHARMACOLOGY - (International Society for Anaesthetic Pharmacology) - The Effect of Rigidity, Shape, Unsaturation, and Length on the Anesthetic Potency of Hydrocarbons - IMPLICATIONS: Molecular flexibility is not a necessary structural requirement for anesthetic action, at least for compounds having six carbon atoms or fewer. Unsaturation, particularly a triple bond, increases anesthetic potency defined either by minimum alveolar anesthetic concentration or the Meyer-Overton constant./

ANESTHETIC PHARMACOLOGY - (International Society for Anaesthetic Pharmacology) - The Effects of Benzodiazepines on Human Opioid Receptor Binding and Function - IMPLICATIONS: Several human and animal studies have shown analgesic effects of benzodiazepines after spinal injection. Our results show that large concentrations of midazolam, chlordiazepoxide, and diazepam displace the binding of (3H)-diprenorphine -- An opiate radioligand from k receptors. In an in vitro functional assay, midazolam is a weak agonist at the d-opioid receptor, whereas all three benzodiazepines are k-Opioid agonists. These findings may partially explain the mechanism of benzodiazepine-induced spinal analgesia./

ANESTHETIC PHARMACOLOGY - (International Society for Anaesthetic Pharmacology) - The Effects of Isoflurane on Native and Chimeric Muscarinic Acetylcholine Receptors: The Role of Protein Kinase C - IMPLICATIONS: By use of the Xenopus oocyte expression system, we investigated the effects of isoflurane on muscarinic signaling and the role of protein kinase C in these effects. Our findings suggest that isoflurane inhibits muscarinic receptors through activation of protein kinase C and that the relevant phosphorylation sites are located outside the third intracellular loop./

ANESTHETIC PHARMACOLOGY - (International Society for Anaesthetic Pharmacology) - The Effects of Low-Flow Sevoflurane and Isoflurane Anesthesia on Renal Function in Patients with Stable Moderate Renal Insufficiency - IMPLICATIONS: The serum creatinine and blood urea nitrogen data indicate that, for exposures of <130 ppm/h in Compound A inspired area under the curve, renal effects of low-flow sevoflurane are similar to those of isoflurane in patients with stable renal insufficiency./

ANESTHETIC PHARMACOLOGY (International Society for Anaesthetic Pharmacology) - The Inhibitory Effect of Bupivacaine on Prostaglandin E2 (EP1) Receptor Functioning: Mechanism of Action - IMPLICATIONS: Clinically relevant bupivacaine concentrations inhibit prostaglandin E2 EP1 subtype signaling. This may explain the effects of regional anesthesia on physiologic responses such as fever, inflammation, and hyperalgesia during the perioperative period./
